The heart is still immature, but the major components, such as the bulbous arteriosus, pro-epicardium, and cardiac valves, are formed. From 48 hpf, the ventricle and the atrium become morphologically distinguishable as two separate chambers. At this stage, the irregular cardiomyocyte contractions became coordinated, with an increase of sequential contractions from the atrium and the ventricle, in relation to cardiac development. After the formation, the tube folds toward the right side, becoming the future ventricle, positioned on the opposite side with respect to the atrium, on the left side. The disc elongation generates a heart tube, that is completely formed at 28 hpf, while the cardiogenic differentiation continues. Zebrafish Cardiovascular System: Morphogenesis and Physiology A detailed scheme of the zebrafish life cycle is reported in Figure 1. At ~3 months, zebrafish are sexually mature and are considered adults. At ~6 weeks, zebrafish change their pigment patterns and their fins gain the characteristic of a juvenile appearance. After 5 days, zebrafish begin to be considered experimental animals. During this phase, the morphogenesis is completed and the larvae lose transparency and develop pigmentations. The hatching time can occur between 48 and 72 hpf. After the formation of the head, fins, circulatory, and other primary systems, at 24 hpf, the heart initiates cardiac contraction. During the stage between 10 and 24 hpf, the body of the embryo increases quickly and the sensory tissue start to form. Embryos develop synchronously and extremely rapidly. A single breeding couple produces hundreds of eggs, where the external fertilization and the transparency of embryos allow us to explore the morphology and function of developing organs by light microscopy. The zebrafish life cycle is distinguished by four main stages: the embryo, until 50 h post-fertilization (hpf) larva, until metamorphosis at ~28 dpf the juvenile, from ~3 months until the end of puberty and the adult stage. Many genes in zebrafish are orthologous to humans, but are present in two copies, creating another molecular pathway with different effects than the main one. The extremely rapid development could be a disadvantage, where the screening in zebrafish appears as a running-away train, requiring a perfect experiment setting to have the right exposure time. The lack of maternal-embryo interaction during gestation and the presence, up to 48–72 hpf, of the chorion, may create problems for drug permeability. Like all animal models, zebrafish also present some disadvantages. This, together with the ease of making transgenic models, provides the generation of gene expression and cell-specific reporter lines to follow real-time in vivo experiments, useful for the study of physiological and pathological conditions. Generally, hundreds of embryos can be easily housed in a standard aquarium, allowing not only a reduction of housing space and testing in reasonable sample sizes but also a very high number of replicates at one time. More specifically, embryonic cardiac development and physiological responses to exposure to numerous toxin substances are similar to those observed in mammals, making zebrafish an ideal model to study cardiac development, toxicity, and cardiovascular disease. The high conservation of gene function, low cost, small size, rapid growth, and easiness of genetic manipulation are just some of the features of this experimental model. Over the last few decades, zebrafish has become a powerful non-mammalian model to study alterations associated with human disease. Mammalian models, sharing structural and functional similarities with humans, have been widely used to study cardiovascular disease mechanisms and test new therapies, but many of them are rather expensive and difficult to manipulate. The gradual decline of the kidney leads to the accumulation of uremic solutes with a negative effect on every organ, especially on the cardiovascular system. Cardiovascular events are the main mortality cause in CKD, with a cardiovascular risk 10 times higher in these patients than the rate observed in healthy subjects. About 10% of the general population is affected by CKD, representing the sixth cause of death in the world. Chronic kidney disease (CKD) is an increasing health care problem.

Finding new life in a play that most people who sit in the audience will have some preconceived notion about? A big, meaty challenge. Keeping the audience interested, even laughing, when there are only two of you onstage for many long minutes? A challenge. That torqueing, circular dialogue? A challenge. With Godot, every part’s the challenging center of attention there are no maids or butlers here. Among our own changing circumstances, it abides. Where Shakespeare’s words are endlessly adapted-cut up, re-worded, placed in new settings such as the controversial production of Julius Caesar at the Public Theatre’s Shakespeare in the Park this summer- Godot does not change to suit us. And although theaters have ignored his dictates, what he wanted is what we most often see: four male actors, the dead tree set, the uncut dialogue, the gloom. Because of Beckett’s demands, productions of Godot rarely change. just some guy named Godot?).Īnd it’s always these same elements, pared to the bone. While the men and their momentary visitors, Pozzo and Lucky, are distinct, nearly everything else is open for interpretation, from the set (post-Apocalyptic or wintertime?) to the costumes (former businessmen gone to ruin or vaudeville performers?) to Godot himself (God or. From our position in the audience, we watch two men, Vladimir and Estragon, and listen to their dismal, circulatory debate about whether Godot will show up and what they should do if he doesn’t. It manages to combine a specific tone and characters with an elusive setting and arc. The play is strong on its own merits, cultural zeitgeist aside. That’s the kind of longevity and cultural impact that most playwrights would kill for. Already this year, Stephen Colbert appropriated it to skewer the health care debate, and Elon Musk named his nearly-impossible-to-build-but-finally-working tunnel driller after it. And the play’s cultural reach is even greater than these production numbers indicate. Samuel French, Inc., which licenses it, reports that Godot will be professionally produced at least ten times around the world in the next three months, nearly 65 years after it first premiered. Why do theaters keep presenting Samuel Beckett’s Waiting for Godot? Somehow, this long, apparently dystopian play has become as perennial as The Music Man. But I digressed and clicked the link, leading to where I could play the game on my browser so I didn’t have to risk downloading it and getting any creepy viruses, I guess. I put my hand over my face and corpsed, thinking this was all going to be another “Lavender Town Syndrome” hoax. The page itself had the story and stated that players who were able to finish this original arcade game board suffered severe psychological and occasionally physiological trauma, up to and including death of the subject (and that the ROM's effect is significantly reduced). I opened the link up and was taken a browser page. However, I was reminded of the link my friend gave me thanks to the trusty Notepad file I saved to my desktop. Honestly, I didn’t believe him, but he gave me a link that I kept noted for a few days later since I was busy with school at the time.Įventually when I got home one late Friday, I hopped on my computer as I was originally going to play more L4D2 or Quake Live. One day, I was chatting with my friend on Skype, who allegedly told me that he found a supposed version of Pac-Man: A ROM that was found in locked away in a safe in the back of an old arcade in some abandoned amusement park and extracted from a damaged PCB board. I originally played the original arcade game on World 2’s “Arcade” bonus area which allowed you to play all of the classic games from the 80’s. Before I start, it’s obvious that I'm a Pac-Man fan. I didn’t like the new reboot they gave him and all, though, as they made him an annoying high school kid trying too hard to be cool. The fella’ will always have a place in my heart since the days I played Pac-Man World 2 on my GameCube. It should be a reasonable default. You remember Pac-Man? Yeah, the little yellow ball who has been the main mascot for Namco (now merged with Bandai) for 41 years now. MSYS2 provides several compiler flavors, UCRT64 being one of them. If it closes itself during the update, restart it and repeat the same command to finish the update. If done correctly, the terminal prompt will say UCRT64 in magenta letters, not MSYS. I myself am not sure about why this happens (I think it has something to do with Sourceforge URL redirection or whatever that the installer currently can't handle properly enough).Īnyways, if you're already planning on using MSYS2, there's no need for that installer.Īfter the install completes, click on the MSYS2 UCRT64 in the Start menu (or C:\msys64\ucrt64.exe). Unfortunately, the MinGW-w64 installer you used sometimes has this issue. There are some advanced settings, such as tunnel IPv6, DNS leak protection, and ports, but unless you know how they work (and what they do), you’re better off leaving the settings alone unless you contact customer support. If you pay for the yearly plan, it’s not as inexpensive as some VPN services, but if you pay by the month, it’s one of the most economical solutions available. Some users may be cautious about using a VPN that looks so “bare bones,” but you have access to relevant information like your connection status, how much time is remaining, and the software version. The Mullvad VPN service stands out for one simple reason: no matter how you pay or for how long you pay, it just costs 5.42 a month. While you will automatically be connected to the best server, you can click on the menu and choose a specific server. That 5EU fee is significantly less than the average monthly price of the VPNs weve. Each account allows up to five simultaneous connections, which is an ideal option for a household or a user with multiple devices. Only a few for-pay VPNs, such as Kaspersky Secure Connection, are cheaper. You can also use the Mullvad OpenVPN to install it on iOS, Android, Chromebook, Linux, and other desktop operating systems. Mullvad VPN has apps for macOS, Windows, Debian, and Fedora. A Mullvad subscription costs 5 per month (approx. You can download the software, and the app will automatically install itself as long as it’s compatible with your device. Mullvad is a provider that has a rather unusual pricing scheme. To get a Mullvad account, all you need to do is register, complete an easy captcha, and then you’ll receive an account number. The company’s three-year plan costs a little less than 108, or just under 36 per year. We understand that our idea of “ease of use” might be different than yours, but Mullvad VPN is one of the more user-friendly VPNs available. Let’s take a look at two of the most popular VPN choicesMullvad and NordVPN. It's not the cheapest option on the market, but it's close, and the 5 /month price has never changed, which probably can't be said for any of its competitors. Most VPNs are designed with usability in mind. When it comes to privacy, it's absolutely one of the best VPNs out there, if not the best. You don’t need to be an experienced tech user to use a VPN. It’s going to be a meaty start, not to mention the last few holes to finish.”Īs ESPN statisticians noted, Spieth is the British Open’s first defending champion since Woods in 2006 to hold at least a share of the lead after 54 holes. The way that it moved throughout the day and the timing that we were playing those holes was ideal. “There were a few holes that obviously didn’t. “We played almost every hole with a little bit of a helping wind today,” Spieth said. The forecast today calls for those breezes to turn to gusts of up to 25 mph, the kind that explain this course’s “Car-nasty” nickname. And Saturday was idyllic, with fluffy clouds and gentle winds.
